What Is Syn-Coll?

Syn-Coll (palmitoyl tripeptide-5) is a lipopeptide cosmetic active developed by DSM that directly mimics TGF-β (Transforming Growth Factor Beta) — a key growth factor in wound healing and collagen production. By activating TGF-β signaling in skin fibroblasts, Syn-Coll stimulates production of collagen types I, III, and IV.

TGF-β is one of the most important signals for collagen synthesis in the skin. As TGF-β signaling declines with age, collagen production drops, leading to wrinkles and skin thinning. Syn-Coll aims to restore this signal topically, essentially telling skin cells to produce collagen as if they were younger.

The palmitoyl (C16 fatty acid) modification enables skin penetration and cellular uptake. Syn-Coll is used in premium anti-aging skincare formulations, often in combination with Matrixyl and other peptide actives.

What The Research Says

In vitro studies demonstrate Syn-Coll activates TGF-β pathway signaling and increases collagen production in human dermal fibroblasts in a dose-dependent manner. Clinical studies showed significant wrinkle depth reduction after 84 days of topical use.

Comparative studies suggest Syn-Coll may be more effective at stimulating collagen than some matrikine peptides due to its direct growth factor mimicry rather than indirect matrikine signaling.
